Abstract Objective: To explore apoptosis of the fibroblast induced by Fas and Fas-L after trabeculectomy. Method: A total of 20 healthy white rabbits were randomly divided into the control group and observation group,10 cases in each group,and the rabbits in the observation group were given glaucoma trabeculectomy.Two groups of tissues in trabeculectomy filtration area, which contain conjunctiva, anadesma, sclera and limbus corneae were cut down, and the fibroblasts of the tissues were cultivated. 5 cases of fibroblasts in each group were randomly selected and treated by FasMcAb. At last, PI staining and flow cytometry were used for testing the apoptotic rate of all the fibroblasts.Result: The apoptotic rate of normal fibroblasts induced by FasMcAb was higher than that of normal fibroblasts without induced by FasMcAb, the difference was statistically significant(P<0.01); the apoptotic rate of fibroblasts from tissues in trabeculectomy filtration area induced by FasMcAb was higher than that of fibroblasts from tissues in trabeculectomy filtration area without induced by FasMcAb,the difference was statistically significant(P<0.01);the effect of FasMcAb on the apoptotic rate of fibroblasts from tissues in trabeculectomy filtration area was more significant than that on normal fibroblasts, the difference was statistically significant(P<0.01).Conclusion:Fas/Fas-L can accelerate apoptosis of the fibroblast after trabeculectomy, and helps to find a new breakthrough to reduce the scar after glaucoma trabeculectomy.
