Abstract Objective: To investigate the clinical application value of mean erythrocyte volume (MCV) and hemoglobin (Hb) electrophoresis in screening for thalassemia in pregnant women during second trimester. Method: The clinical data of 320 second trimester pregnant women who underwent pregnancy screening in Ningde Municipal Hospital of Ningde Normal University from May 2019 and June 2021 were retrospectively analyzed, and 124 thalassemia patients were classified as the thalassemia group and 196 non-thalassemia patients were classified as the non-thalassemia group according to the genetic test results, and both groups received MCV and Hb electrophoresis, and the test results of the two groups were compared. Result: The positive rates of MCV and Hb electrophoresis in the thalassemia group were higher than those in the non-thalassemia group, and the differences were statistically significant (P<0.05). The sensitivity of MCV and Hb electrophoresis for the diagnosis of thalassemia were 87.10% and 83.06%, the specificity was 70.92% and 72.45%, and the accuracy was 77.19% and 76.56%, respectively. The sensitivity, specificity and accuracy of the combined assay were 97.58%, 94.39% and 95.63%, respectively. The sensitivity and specificity of the combined test were higher than those of the single test, and the differences were statistically significant (P<0.05). Conclusion: MCV and Hb electrophoresis have high sensitivity and specificity in screening and diagnosis of thalassemia in pregnant women in the second trimester, and the combined test can improve the diagnosis rate.
