The Protective Effect of Calpain Inhibitor Against Focal Cerebral Ischemia-reperfusion Injury in Rats |

Abstract Objective: To study the protective effects of calpain inhibitor calpeptin on the ischemic/reperfusion damage to the brain with the model of left middle cerebral artery ry occlution-reperfusion( MCAO) in rats. Method: Rats were randomly divided into four equal groups: group calpeptin, vector dimethyl sulfoxide( group DMSO), ischemic/reperfusion group( group MCAO), and sham operation group( group sham). After 2 hours of left middle cerebral artery(MCA) occlusion, followed by 12, 24 and 48 h of recirculation. After awakening from anesthesia, neurological functions of rats were evaluated with the 5 grade scale developed by Longa, Infarct zones were confirmed by TTCstaining, TUNEL was used to detect neuronal apoptosis in hippocampal CA1 section. Result: Compared with group MCAO rats, neurological functions and neuronal apoptosis of hippocampal CA1 section and infarct size of group DMSO had no significant difference.Neurological functions of group calpeptin had significant improvement; brain infarct size and neuron apoptosis of hippocampal CA1 section of group calpeptin were significant reduced. Conclusion: Calpeptin could significantly protect the brain against the ischemic/reperfusion damage in rats.
