Abstract Objective：To extract human genomic DNA with HBV infection，and preparation of DNA template for studying the genetic polymorphism of HBV infection.Method：HBV infection of human anticoagulant whole blood were collected，Human genomic DNA was extracted by Miller’s Method.Result：DNA template in HBV infected person included asymptomatic carrier，chronic hepatitis，severe hepatitis，liver cirrhosis and hepatocellular carcinoma，and part of a healthy blood donors were collected at the same time.Conclusion： The template library is mainly HBV persistent infection，extracted DNA with Miller’s Method has high purity and stability.
