Abstract Objective:To investigate the protective effect of ginkgolide B on retinal ganglion cells after crush of intra orbital optic nerve in rats.Method:Thirty-six SD rats which were 46-day-old were randomly divided into noral control group(A group,12 rats),model control group(B group,12 rats)and Ginkgolide B group(C group,12 rats). In each group, only the right eyes were used.No operation was carried out in normal control group, corresponding volume of NS or Ginkgolide B at 40 mg/kg was injected intraperitoneally into the rats from one week before crush until one week after crush in model control group or Ginkgolide
B group, one week and two weeks after operation, flash visual evoked potential were performed. Two weeks after operation, calculated the number of RGCs in each group.Result:one week after operation, the coefficients of LPl and APl were[(90±4)ms,(19±3)μV],[(110±6)ms,(11±3)μV]and[(94±4)ms,(18±2)μV]respectively in noral control group, model control group and Ginkgolide B group, the differences were statistically significant among B group and A group or C group(P<0.05).Two weeks after operation, the coefficients of LPl and APl were[(92±11)ms,(21±3)μV],[(186±15)ms,(7±2)μV]and[(104±12)ms,(14±3)μV] respectively, the number of RGCs were(281±39),(112±20)and(228±35)respectively,the differences were statistically significant in 3 groups(P<0.05).Conclusion:Ginkgolide B can partly inhibit the apoptosis of RGCs and promote the survival of RGCs after crush of intra orbital optic nerve.
