Abstract Three-dimensional cell culture in vitro can not only simulate the microenvironment of intercellularsignal transduction between cells and extracellular matrix， but also can reproduce the conditions in cell culture. In recentyears， three-dimensional cell culture technology has increasingly widely used in biomedical research， which becomesa powerful tool for drug toxicity assay， tumor multi-drug resistance， high-throughput screening of anticancer drugs anddrug metabolism. In this paper， we give a brief review of their latest research progress on construction of three-dimensionaltumor spheroid in vitro.
