Abstract Objective:To explore the expression of long chain of non-encoded RNA-HOTAIR in sentinel lymphnode and axillary lymph node in breast cancer, to analyze the correlation with other indicators of breast cancer, and tohelp guide treatment and predict outcome. Method: Data from 10 surgical patients of breast cancer hospitalized from June2013 to December 2013 was collected. Sentinel lymph node biopsy was performed, and breast tumor tissue, paracanceroustissue,Sentinel lymph node(SLN),and axillary lymph nodes( ALN) were collected.Invasive ductal carcinoma wasconfirmed by pathological examination.Expression of HOTAIR in 4 sets of specimen tissue was measured by real-timefluorescence quantitative RT-PCR.Combined with clinical pathology, the correlation with other indicators was evaluatedusing t test and rank correlation.Result:(1)HOTAIR gene expression in each group was measured.(9.33±1.23) forSLN group and( 6.90±2.00) for ALN Group( t=3.401,P=0.004).(8.81±1.27) for Cancer Group, and( 7.00±1.11)for paracancerous tissue group( t=3.389,P=0.003).The difference of the expression of HOTAIR between SLN group andthe adjacent tissue Group was statistically significant(t=4.441,P<0.001).(2)HOTAIR expression was correlatedwith Clinical pathological indicators. It was positively correlated with Ki-67 protein expression and lymph node metastasis
rate, and the correlation coefficient of 0.67 and 0.82, respectively(P<0.05).Conclusion:Abnormal high expressionof HOTAIR was observed in the Sentinel Lymph nodes of breast cancer as well as the tumor tissue.It can promote cellproliferation, invasion and metastasis,which may provide new therapeutic target for breast cancer
