Objective:To investigate the role of ganoderma lucidum spore oil on tumor cell proliferation, androgen receptor activation and transcriptional activity as well as related kinase activation in prostate LNCaP cancer cells. Method: The effects of ganoderma spore oil on the LNCaP cells of the prostate cancer cells were studied by using plasmid transfection,Western blot,CCK-8 cell proliferation assay and dual luciferase reporter gene activity.Result:LNCaP cancer cells proliferation induced by DHT was significantly reduced by ganoderma lucidum spore oil. At the same time, the Western blot assay showed that ganoderma spore oil decreased the phosphorylation activity of AR and protein kinase D in prostate cancer LNCaP cells, but had no effect on the expression of the protein kinase DHT. Luciferase reporter gene activity assay indicated that the activity of AR in LNCaP cells induced by DHT was significantly inhibited by Ganoderma spore oil.Conclusion:Ganoderma lucidum spore oil may contributes to LNCaP cancer cells growth inhibition through down-regulation of AR activation and transcriptional activity in androgen dependent prostate cancer cells.
